• Brain Concentrations-

  When 7-OH was generated as a metabolite of Mitragynine, its brain concentration was significantly lower than when it was delivered alone, even at equal effective doses for analgesia. In particular, as a metabolite of Mitragynine, the concentration of 7-OH in the brain was 11 times lower.

• Contribution To Mitragynine’s Effects- 

  Research indicates that the amount of 7-OH generated as a metabolite of Mitragynine is sufficient to explain its opioid-mediated analgesic effect, even though it has a higher potency. In mice, the parent chemical mitragynine does not directly impact the analgesic effect of the molecule.

• Complex Pharmacology- 

  Mitragynine has a complicated pharmacology that combines activity at non-opioid receptors with low-efficacious mu-opioid receptor agonism. However, 7-OH seems to be a more effective and selective mu-opioid receptor agonist.

• Metabolic Relationship- 

  One way that Mitragynine works in the body is by converting it to 7-OH. A minor mechanism that results in the conversion of 7-OH back to Mitragynine is also possible.
